Pyrogen test is performed to check the presence or absence of pyrogens in all aqueous parenterals. Rabbits are used to perform the test because their body temperature increases when pyrogen is introduced by the parenteral route.

What is the purpose of pyrogen?

The pyrogens inhibit heat-sensing neurons and excite cold-sensing ones, and the altering of these temperature sensors deceives the hypothalamus into thinking the body is cooler than it actually is. In response, the hypothalamus raises the body's temperature above the normal range, thereby causing a fever.

What does the pyrogen test measure?

The test methods measure cytokine levels (either IL-1-beta or IL-6) from human blood cells or a human monocytoid cell line. Increased cytokine release is used as a biomarker of a pyrogenic response.

What is the principle of pyrogen test?

The pyrogen test on rabbits is based on the measurement of the increase in the rabbit's temperature upon being injected with a product that could contain a contaminant of the pyrogen type. The pyrogens, as their name suggests, refer to all the substances that cause an increase in fever, also known as pyrexia.

What are pyrogens explain pyrogen testing in detail?

A pyrogen is a foreign substance that causes a fever (temperature elevation) in an animal's body. Typically, pyrogenic substances include endotoxin and other bacterial byproducts. Vaccines and other injectable drugs must be confirmed to be pyrogen free according to regulatory requirements of 21CFR, USP, and EP.

Why is Lal important?

The LAL (limulus amebocyte lysate) testing, also known as bacterial endotoxin testing, is an in vitro assay used to detect the presence and concentration of bacterial endotoxins in drugs and biological products, and is an important part of pharmaceutical microbiology.

Which animal is used in pyrogen test?

Animal Tests

In the rabbit pyrogen test (RPT), which has been in use since the 1940s, rabbits are restrained and injected with a test substance while their body temperature is monitored for changes that suggest the substance might be contaminated with pyrogens.

What are pyrogens made of?

Pyrogens are fever-inducing substances usually derived from microorganisms [endotoxins or lipopolysaccharide (LPS)] and when present systemically in sufficient quantity can lead to severe signs of inflammation, shock, multiorgan failure, and sometimes even death in humans.

How is LAL test performed?

Test procedure: A BET involves analyzing the liquid sample or sample extract using Limulus Amebocyte Lysate (LAL). LAL is a reagent made from the blood of the horseshoe crab. In the presence of bacterial endotoxins, the lysate reacts to form a clot or cause a color change depending on the technique.

What do you mean by the term pyrogen?

: a fever-producing substance.

What is LAL test of pyrogen?

Limulus amebocyte lysate test is an aqueous extract of blood cells (amoebocytes) which obtain from the horseshoe crab ( Limulus polyphemus ). ... LAL test is recommended in all international pharmacopeias as the method for finding bacterial endotoxins. Gram-negative bacteria produce endotoxins (pyrogen).

What is detected in pyrogen testing of pharma product?

Learn how to determine the Pyrogen in injection samples and water for injection using animals. The test involves measurement of the rise in body temperature of rabbits following the intravenous injection of a sterile solution of the substance under examination.

What is the significance of pyrogen in pharmaceutical preparations?

RADIOCHEMICAL METHODS | Pharmaceutical Applications

Bacterial endotoxins (pyrogens) are polysaccharides from bacterial membranes. They are water soluble, heat stable, and filterable. If they are present in a preparation and administered to a patient they can cause fever and also leukopenia in immunosuppressed patients.

Why do pyrogens cause fever?

Fevers are caused by chemicals called pyrogens flowing in the bloodstream. Pyrogens make their way to the hypothalamus in the brain, which is in charge of regulating body temperature. When pyrogens bind to certain receptors in the hypothalamus, body temperature rises.

What is pyrogen reaction?

Pyrogen reaction is a febrile phenomenon caused by infusion of solution contaminated, and commonly manifested by cold, chill and fever [1]. With improved sterilization and generalized application of infusion set (single-use), the prevalence of pyrogen reaction has been controlled, but still exists in clinical practice.

What cells produce pyrogens?

The endogenous pyrogens are produced by the immune cells that are activated by the presence of infectious agents (e.g. bacteria, viruses, prions, and viroids). Endogenous pyrogens are usually cytokines, such as interleukin-6, interleukin-1, tumour necrosis factor, interferon-alpha, gp130 Receptor Ligands, and so on.

Why LAL test is done?

The LAL test is carried out as a method for the control of pyrogens at all stages of the production of medicines and biological products. ... The clinical tests conducted with the goal of determining the presence of endotoxins also use the LAL test as the method for the detection of endotoxins.

What is pyrogen chemically?

Pyrogens are substances that can produce a fever. The most common pyrogens are endotoxins, which are lipopolysaccharides (LPS) produced by Gram-negative bacteria such as E. coli. The limulus amoebocyte lysate (LAL) test is used to detect endotoxins.

Why is bacterial endotoxin test important?

Bacterial endotoxin testing (BET) is a fundamental safety requirement in the pharmaceutical and biomedical industries. If bacterial endotoxins enter patients' bloodstream in sufficient concentrations, this can cause harmful symptoms such as fever and septic shock and can be fatal in the most severe cases.

What are the types of pyrogens?

There are two types of natural pyrogens: (1) endogenous pyrogens that the host's pyrogen cytokines and (2) exogenous pyrogens that are microbial substance (e.g. lipopolysaccharides in the cell wall of certain bacteria).

What effects do pyrogens have in the body?

When bacterial pyrogens are injected in sufficient amounts, perhaps in microgram quantities, the fever produced is accompanied by chills, body aches, a rise in blood pressure, and possibly a state of shock and death.

What statement best describes the function of pyrogens?

What statement best describes pyrogens? Pyrogens increase secretion of thymosin. Pyrogens cause the hypothalamic thermostat to reset to a higher range. Pyrogens interfere with the ability of bacteria or viruses to infect other cells.

What is the rabbit pyrogen test?

pyrogen test was first published in 1986. The test involves measuring changes in the body temperature of rabbits after intravenous injection of a sterile solution of the solution needing examination. ... As a result, a test covering all types of pyrogens is required to confirm the absence of non-endotoxin pyrogens.

What is pyrogen in pharmaceutical industry?

A pyrogen is a substance that causes fever after intravenous administration or inhalation. Gram negative endotoxins are the most important pyrogens to pharmaceutical laboratories.

Why pyrogens are required to be removed from parenteral products?

Pyrogens can often be difficult to remove from solution due to the high variability of their molecular weight. Pyrogens are also relatively thermally stable and insensitive to pH changes. However, several removal techniques exist. Endotoxins are negatively charged, and will bind to an anion exchanger.
