32P labelling. For use in autoradiography, a radioactively-labelled dNTP molecule is synthesized with the first ("alpha") PO ₄ containing radioactive ³²P rather than non-radioactive ³³P.

Which of the following reactions is used to radioactively label DNA?

Nick translation is a technique for radioactively labeling double-stranded DNA, making it suitable as a hybridization probe for detecting specific genomic sequences.

How do you radioactively label DNA?

To radioactively label a DNA fragment for use as a probe, one of the incorporated nucleotides provided in the reaction is radiolabeled on the alpha phosphate position. The translated nick can be sealed by DNA ligase.

What is 32P labeling?

32P-postlabeling analysis is an ultrasensitive method for the detection and quantitation of carcinogen-DNA adducts. ... It has a wide range of applications in human, animal and in vitro studies, and can be used for a wide variety of classes of compound and for the detection of adducts formed by complex mixtures.

Which kinase is used to label the DNA?

The Invitrogen KinaseMax 5' End-Labeling Kit allows the efficient end-labeling of DNA or RNA to high specific activity with T4 polynucleotide kinase and [gamma-³²P] ATP, or quantitative phosphorylation of 5' ends using unlabeled ATP.

What enzymes are used to make radioactively labeled DNA?

The nick translation reaction is used to introduce radioactive nucleotide phosphates into unlabeled DNA for the purpose of making a probe. The reaction depends on the ability of the enzyme DNA polymerase I to initiate DNA synthesis at free 3´ OH groups, which are exposed as nicks in the unlabeled DNA.

What radioactive molecule is used to end label and oligonucleotide?

Radioactive phosphate is commonly added to the 5' or 3' end of an oligonucleotide post synthesis using enzyme-catalyzed reactions.

What is the 32 in phosphorus 32?

Phosphorus 32 (P-32) is the phosphorus isotope whose nucleus consists of 15 protons and 17 neutrons. It disintegrates by emitting a β- (1.71 MeV) particle in 32S with a half-life of 14.263 days. It is an artificial radioactive substance obtained by neutron bombardment of stable phosphorus.

What macromolecule is sulfur 35 labeling?

Phosphorus is not present in most proteins. They used ³⁵S to label proteins. Proteins contain some sulfur, because sulfur is found in the amino acids cysteine and methionine.

What is phosphorus 32 used for?

A radioactive form of the element phosphorus. It is used in the laboratory to label DNA and proteins. It has also been used to treat a blood disorder called polycythemia vera and certain types of leukemia, but it is not commonly used anymore.

Why is it important that one of the dNTPs is radioactively Labelled?

One or more of the dNTPs are radioactively labeled to help visualize the replication products. Finally, each tube gets one of four special nucleotides called dideoxynucleotides (ddNTPs). ... These nucleotides are recognized and used by DNA polymerase, and can be incorporated at the 3' end of a growing chain.

Which type of nucleotide would you use to Radiolabel DNA and RNA?

Radioisotope labeling: Considered as a conventional method for nucleic acid labeling, radiolabeled nucleotides are synthesized using ATP-gamma-³²P or ³⁵P. They are easily incorporated into nucleic acid sequences by traditional enzymatic means or by an organism of interest.

When radiolabeling oligonucleotides with polynucleotide kinase and ATP Where is the label on this ATP molecule?

Oligos can be labeled at either the 3' or the 5' end. Using polynucleotide kinase and ATP (γ-32P), the 5' end is labeled. Using terminal transferase and deoxynucleotide triphosphate the 3' end is labeled.

Which of the following can be used to determine the precise amount of radioactively labeled DNA?

Autoradiography is used for detecting radioactively-labeled DNA or RNA in a gel after separation by electrophoresis. Electrophoresis gels containing RNA or DNA are transferred onto membranes by blotting to allow more convenient handling during autoradiography, or the gel can be dried onto filter paper.

How do you label one end of DNA?

There are two ways to label a DNA molecular; by the ends or all along the molecule. End labeling can be performed at the 3'- or 5'-end. Labeling at the 3' end is performed by filling 3'-end recessed ends with a mixture or labeled and unlabeled dNTPs using Klenow or T4 DNA polymerases.

Which form of DNA testing uses a single stranded piece of DNA that is usually labeled with a radioisotope?

RNA probes are stretches of single-stranded RNA used to detect the presence of complementary nucleic acid sequences (target sequences) by hybridization. RNA probes are usually labeled, for example with radioisotopes, epitopes, biotin or fluorophores to enable their detection.

Why is sulfur used to label proteins?

Radioactive sulfur-35 was used to label the protein sections of the T2 phage, because sulfur is contained in protein but not DNA.

Does protein contain phosphorus?

Proteins may be defined as compounds of high molar mass consisting largely or entirely of chains of amino acids. ... In addition to carbon, hydrogen, and oxygen atoms, all proteins contain nitrogen and sulfur atoms, and many also contain phosphorus atoms and traces of other elements.

Which answer best explains why radioactive sulfur and phosphorus were used to label bacteriophages in the Hershey and Chase experiments?

In their experiment Hershey and Chase labeled the bacteriophages with radioactive isotopes to see where the virus attacks. They use phosphorus 32 and sulfur 35 because proteins contain almost no phosphors and DNA contains almost no sulfur. ... DNA is a deoxyribonucleic acid that holds the code on how to make proteins.

What do we use carbon 14 for?

The isotope also is used as a tracer in following the course of particular carbon atoms through chemical or biological transformations. In carbon-14 dating, measurements of the amount of carbon-14 present in an archaeological specimen, such as a tree, are used to estimate the specimen's age.

How is CA 47 used?

Calcium-47 is used in medicine to investigate bone metabolism problems or to diagnose calcium disorders. It is also used in the biomedical research of animals to study the cellular body function and the formation of bones in mammals.

What is the use of sodium 24?

Sodium 24 is used as an electrolyte tracer to follow the path sodium takes in a person's body to see if their uptake levels are within normal ranges, while sodium 22 is used in nuclear medicine imaging for positron emission tomography. Sodium -24 can also be used in non-medical applications.

What is used to label oligonucleotide probes?

Oligonucleotide probes are often prepared with covalent attachment of a reporter molecule (eg, a fluorescent dye) or affinity labels that allow them to be attached to solid supports. Probes used in homogeneous (real-time) PCR are usually oligonucleotides with a fluorescent label.

Why is probe Labelled?

A probe is a piece of DNA identical (or very similar) to a sequence of interest. In order to locate a specific DNA sequence by hybridization, the probe is labeled with a reporter group. The Klenow fragment of E. coli DNA polymerase is used to make a labeled probe.

Why is a radioactive isotope of nitrogen appropriate for labeling DNA what component of a nucleotide would be labeled?

They decided to label the nitrogen of the DNA, rather than the phosphate. They reasoned that each nucleotide as only one phosphate and two to five nitrogens. Thus, labeling the nitrogens would provide a stronger signal than labeling the phosphates.
