The counterstain is Gram Safranin. The decolorizer is Ethanol. It is added to chemically change the shape of the dye molecule and trap it in the cell wall. Iodine is used in the Gram stain.

What is the Decolorizer in the Gram stain?

The decolorizer, ethyl alcohol, is the most crtitical step. Ethyl alcohol is a nonpolar solvent, and thus penetrates the cell walls of Gram negative cells more readily and removes the crystal violet-iodine complex.

What is the purpose of the Decolorizer in the Gram staining procedure?

Decolorizing the cell causes this thick cell wall to dehydrate and shrink, which closes the pores in the cell wall and prevents the stain from exiting the cell. At the end of the gram staining procedure, gram positive cells will be stained a purplish-blue color.

What is used to Decolorize in the Gram stain method quizlet?

Decolorizer: In order to remove the primary dye from the Gram-negative cells, 95% ethanol is used as a decolorizer. ... Safranin is the secondary dye used in the Gram stain. This dye will stain the decolorized Gram-negative cells, which will appear in pink.

How and why is the Decolorizer the most important step in the Gram stain?

The decolorizing mixture dehydrates cell wall, and serves as a solvent to rinse out the dye-iodine complex. In Gram-negative bacteria it also dissolves the outer membrane of the gram-negative cell wall aiding in the release of the dye.

What happens to the gram positive cell wall during decolorization?

What happens to the Gram-positive cell wall during decolorization? The decolorizing agent dehydrates the peptidoglycan..... Removing water from or dehydrating the peptidoglycan allows the decolorizing agent to shrink the spaces through which the crystal violet-iodine complexes might be able to pass.

How do you Decolorize a Gram stain?

Add Gram's iodine for 1 minute- this is a mordant, or an agent that fixes the crystal violet to the bacterial cell wall. Rinse sample/slide with acetone or alcohol for ~3 seconds and rinse with a gentle stream of water. The alcohol will decolorize the sample if it is Gram negative, removing the crystal violet.

What is the Decolorizer used in the acid-fast stain?

Remel TB Decolorizer is a reagent recommended for use with the Kinyoun or Ziehl-Neelson carbolfuchsin staining procedure to differentiate acid-fast bacteria from nonacid-fast bacteria.

What is the Decolorizer for the acid-fast procedure?

Principle of Acid-Fast Stain

Then the smear is decolorized with decolorizing agent (3% HCL in 95% alcohol) but the acid fast cells are resistant due to the presence of large amount of lipoidal material in their cell wall which prevents the penetration of decolorizing solution.

What is the difference between the Decolorizer of the Gram stain and the acid-fast stain?

The main difference between Gram stain and acid fast stain is that Gram stain helps to distinguish bacteria with different types of cell walls whereas acid-fast stain helps to distinguish Gram-positive bacteria with waxy mycolic acids in their cell walls.

What is the Decolorizer in the Gram stain and why is it important quizlet?

The counterstain is Gram Safranin. The decolorizer is Ethanol. ... Decolorization is important because it will rinse the color out of the gram negative cells so they can be dyed with a secondary dye so you can see the difference between the Gram positive and Gram negative cells.

What is the purpose of the alcohol solution in the Gram stain?

The alcohol dissolves lipids found in the outer cell membrane of Gram-negative bacteria, allowing the crystal violet-iodine complex to leak out of the thinner peptidoglycan layer.

What is Endospore staining in microbiology?

Endospores staining is the type of staining to recognize the presence spore in bacterial vegetative cells. The bacterial endospores need a staining which can penetrate wall thickness of spore bacteria. A method of endospores staining is Schaeffer Fulton method that used Malachite Green.

Is Murein a peptidoglycan?

Peptidoglycan or murein is a polymer consisting of sugars and amino acids that forms a mesh-like layer outside the plasma membrane of most bacteria, forming the cell wall. ... Peptidoglycan is also involved in binary fission during bacterial cell reproduction.

How many alcohol steps are there in the Gram stain?

The performance of the Gram Stain on any sample requires four basic steps that include applying a primary stain (crystal violet) to a heat-fixed smear, followed by the addition of a mordant (Gram's Iodine), rapid decolorization with alcohol, acetone, or a mixture of alcohol and acetone and lastly, counterstaining with ...

What is Mycolic acid in Gram staining?

From Wikipedia, the free encyclopedia. Mycolic acids are long fatty acids found in the cell walls of the Mycolata taxon, a group of bacteria that includes Mycobacterium tuberculosis, the causative agent of the disease tuberculosis. They form the major component of the cell wall of mycolata species.

What is acid alcohol Decolorizer?

Description. Acid alcohol decolorizer is used to differentiate dyes in various staining methods such as the AFB and regressive H&E staining methods.

Which of the following is used as a Decolorizer for the Endospore staining technique?

Here, water is used as a decolorizer. Because malachite green binds relatively weakly, it can be washed off easily. However, it cannot be washed off easily once it is locked in the spore wall. Once they take in the dye, endospores retain the dye and will be resistant to de-staining.

What is a flagella stain?

The flagella stain allows observation of bacterial flagella under the light microscope. Bacterial flagella are normally too thin to be seen under such conditions. The flagella stains employs a mordant to coat the flagella with stain until they are thick enough to be seen. ... Flagella can vary by number and location.

Which of the following is used as counter stain in Gram staining?

For instance, in Gram staining, two dyes are used: the Crystal Violet and Safranin. The Safranin is the counterstain used in this method.

How do you make acid ethanol?

Acid alcohol: Mix 2.0 mL concentrated hydrochloric acid and 98.0 mL 95% ethyl alcohol. 3. Methylene blue: Prepare a saturated solution of methylene blue by adding 1.5 g powdered methy- lene blue to 100 mL 95% ethyl alcohol. Slowly add the alcohol to dissolve the powder.

What are the reagents of a gram stain?

Reagents needed for Gram staining include:

Crystal violet (primary stain) [1] Gram's iodine solution (the mordant) [1] Acetone/ethanol (50:50 v:v) (the decolorizer) [1] 0.1% basic fuchsin solution (the counterstain) [1]

Which step in the Gram stain is the critical step?

The length of decolorization is a critical step in gram staining as prolonged exposure to a decolorizing agent can remove all the stains from both types of bacteria. The final step in gram staining is to use basic fuchsin stain to give decolorized gram-negative bacteria pink color for easier identification.

How is Gram stain prepared?

Gram staining technique requires simultaneous use of chemical reagents for a fixed period followed by washing; Primary stain (crystal violet), Mordant (iodine), Decolorizer (ethanol or acid-alcohol), and Counterstain (safranin or dilute carbol-fuchsin). ...

Why do gram negative bacteria lose the primary stain during decolorization with ethanol quizlet?

What happened when gram negative bacteria is decolorized? Ethanol dissolves the lipids in the outer membrane. Without lipids and with only a thin peptidoglycan layer, the crystal-iodine complex can escape from cell. so the bacteria are no longer stained (transparent).
